  EXCLUSION ASSAY

​​

  • The exclusion assay allows testing the general  anti-microbial properties of your product against the targeted pathogenic microorganisms and/or viruses.

  • A reduction of pathogen adhesion to the host cells indicates an anti-microbial effect, which may be attributed to various mechanisms including i) direct effect on the pathogen, ii) direct binding and neutralization of the pathogen, iii) blocking of specific binding sites used by the pathogen, iv) modulation of the expression of the binding sites on the surface of the host’s cells v) induction of anti-microbial substances production by host cells.

  • A broad range of human cell lines (gut, vagina, gastric etc) are available.

  OVERLAY ASSAY

  • The overlay agar method allows identifying anti-microbial properties of probiotic strains independent of their mechanism of action.

  • in this method, colonies of the probiotic strains on agar plate are covered with a layer of agar containing the pathogen (overlay), enabling direct contact between the probiotic strain and the pathogen. Anti-microbial activity is revealed by an inhibition zone around the colonies of the probiotics.

  AGGREGATION ASSAY

  • Your product may hinder pathogen colonization through direct aggregation of the pathogen.

  • By inducing pathogen aggregation, your product may limit the pathogen’s ability to bind to host cell.

  • Aggregation can be assessed by measuring optical density and/or through microscopic observations.

  CO-CULTRUE ASSAY​

​​

  • The co-culture assay allows identifying pathogen specific growth inhibition properties of your product.

  • Any product may induce modifications of the microenvironment and/or of the protein expression of the targeted pathogens, resulting in inhibiting their growth.

  • Different growth conditions that mimic in vivo conditions can by be tested.

  • The anti-growth effect is assessed by measuring the optical density and determining colony forming units.

  • Your product may directly or indirectly affect the growth of a pathogen. Indeed, many product can induce microbiome modulation, leading to the release of antimicrobial substances by the microbiome.

  AGAR DIFFUSION TEST

  • The agar diffusion assay allows specific identification of anti-microbial properties related to the released compounds (bacteriocins) or modification of the microenvironment.

  • Probiotic strains may be cultured under different conditions, the anti-microbial properties of culture supernatants are tested by means of hole or disk diffusion assays.
